After syncing you can relax as the number of times you connect to the system the data will be backed up, even if you choose to update the software on iPhone the back up will be taken. If you want to restore the software on iPhone, you will be prompted to back up your data by iTunes.
But there would be a question arising in your mind what if the back up on the system fails, what can one do in that case. There is no need to worry about it, firstly the time machine will take care of your system and in the worst case if every thing fails then there are so many mac data recovery software available in the market which will help you recover your crucial data.
